Dosage
Oral. Adultos o niños: 0.15 a 0.30 mg/kg de peso corporal/día, fraccionar en dos tomas, administrar 8 a 12 horas después de suspender la vía intravenosa. Intravenosa. Adultos o niños: 0.05 a 0.1 mg/kg de peso corporal/ día, 6 horas después del transplante.
General information
Macrólido inmunosupresor que inhibe la activación del linfocito-T al unirse a una proteína intracelular FKBP-12 bloqueando la actividad de calcineurina, calmodulina y calcio, evitando la generación del factor nuclear de células T activadas.
Pregnancy warnings
Son evidentes las anormalidades en fetos humanos. Deben prevalecer los factores de riesgo, sobre los pretendidos “beneficios” del fármaco a prescribir. Contraindicado en el embarazo.
Precautions
Puede aumentar la susceptibilidad a las infecciones, riesgo para desarrollo de linfoma.
Adverse effects
Cefalea, temblor, insomnio, diarrea, náusea, anorexia, hipertensión arterial, Hiperpotasemia o hipokalemia, hiperglucemia, hipomagnesemia, anemia, leucocitosis, dolor abdominal y lumbar, edema periférico, derrame pleural, atelectasia, prurito, exantema, nefropatía tóxica.
